En 1950, la Ville de Nice acquiert la villa Garin de Cocconato, également appelée villa des Arènes, à deux pas de la résidence du maître, au Regina. L'idée fait son chemin d’en faire un lieu où seraient présentées à la fois les donations faites par le peintre lui-même ainsi que celles de ses héritiers. Créé en 1963, le musée Matisse prend alors place au premier étage de la villa, le rez-de-chaussée étant occupé par le musée d'Archéologie. En 1978, suite à la donation Jean Matisse, la Ville de Nice veut procéder au réaménagement du musée afin de mieux mettre en valeur les collections tout en préservant le caractère intimiste de la donation initiale de Madame Henri Matisse. La complexité des procédures, les travaux de fouilles sur le futur site et les recours, ralentirent considérablement la construction d’un nouveau musée d’archéologie qui ne fut achevé qu’en 1989. Le déménagement de ce-dernier sur le site romain voisin permet au Musée Matisse d’occuper la totalité de la villa. Le musée Matisse fait l'objet d'un programme de rénovation et d'extension. Il accueille aujourd'hui la collection permanente constituée de diverses donations, celle du peintre lui-même qui vécut à Nice de 1917 à 1954, de ses héritiers ou mises en dépôt par l’État.
