Le musée, est créé à l'initiative de la Société historique et archéologique de Château-Thierry, cession du 18 décembre 1876. Il s'enrichit ensuite de collections autour de Jean de La Fontaine et de son œuvre, grâce à de riches donateurs, passionnés par l'homme de lettres. Une politique d'acquisition, menée avec l'aide de la Direction des musées de France, du Fonds d'acquisition pour les musées de Picardie (FRAM), de la Ville et du mécénat local, permet l'acquisition de pièces uniques, telle l'importante collection du baron Feuillet de Conches. Les collections s'articulent autour de l’œuvre du poète qui a inspiré une importante production artistique du XVIIe siècle à nos jours, en France et dans le monde. Le musée conserve aussi une petite collection de beaux-arts (de qualité variable) rassemblée au XIXe siècle alors que l'orientation de la politique d'enrichissement autour de la figure de Jean de La Fontaine n'était pas encore clairement définie.
