Les collections du musée ont une double origine : les collections du musée Calvet d'Avignon et la collection Campana. Les pièces provenant du musée Calvet (une soixantaine de peintures et 600 pièces de sculpture) représentent l'ensemble des œuvres du Moyen Âge et de la Renaissance (assemblées depuis la création de la fondation du docteur Esprit Calvet en 1810 jusqu'à nos jours). La grande majorité des peintures provient de la réunion d'une partie de la collection Campana, répartie par l’État dans divers musées de province entre 1853 et 1876.
