Le musée est fondé en 1855 par Pierre Eudoxe Dubalen. Les collections sont alors orientées vers l'histoire naturelle. Les familles et les élèves de Charles Despiau et Robert Wlérick constituent le premier soutien pour la création d'un musée dédié à leur mémoire dans les années 1950. Il est transféré de l'Hôtel de Ville à la Tour Lacataye, restaurée en 1963 et inaugurée en 1968, sous la double impulsion du Maire, Charles Lamarque-Cando et d'un industriel montois, amateur d'art et collectionneur, Raymond Farbos. Ce dernier préside les "Amis de Charles Despiau et Robert Wlérick", association chargée d'animer l'institution par des expositions orientées sur l'art moderne et contemporain. Dans les années 1980 et19 90, le musée se spécialise dans la promotion de la sculpture figurative de l'entre-deux guerres et fait de Mont-de-Marsan la ville de la sculpture. Les salles d'expositions des collections permanentes ne sont plus accessibles. Le musée se prépare à des travaux de rénovation jusqu'à fin 2024.
