Dès la Révolution, un cabinet d'histoire naturelle et quelques œuvres d'art forment le noyau du musée. En 1857, grâce à l'impulsion donnée par Léon de La Sicotière, sénateur et érudit local, les collections de beaux-arts sont développées (donations Chennevières, His de la Salle, Leriche, Jacquette, Noblesse...). Le musée est installé à l'hôtel de ville, puis à la maison d'Ozé. Les dentelles commencent à entrer dans les collections au début du XXe siècle, comme témoins de l'industrie locale. En 1981, l'ensemble du patrimoine muséal alençonnais est regroupé dans l'ancien collège des Jésuites.
