Le musée trouve son origine en 1837, avec l'achat par la ville de Cassel d'un cabinet d'histoire naturelle. Dans les années 1950, le musée se consacre à la mémoire du maréchal Foch et de la Grande Guerre, Cassel ayant accueilli le quartier général de Ferdinand Foch, entre octobre 1914 et juin 1915. En 1964, est inauguré le musée d'art, d'histoire et de folklore dans l'Hôtel de la Noble-Cour. En 1997, le musée de Cassel passe sous tutelle du Département. S'ouvre alors une longue période durant laquelle se construit le projet scientifique et culturel. Les collections sont enrichies grâce à une importante politique d'acquisition. Après l'inauguration des réserves dans un bâtiment neuf spécialement conçu pour la conservation des œuvres, en 2009, le musée départemental de Flandre rouvre ses portes en octobre 2010, après deux ans de travaux.
