En 1810, Esprit Calvet lègue à la ville d'Avignon son cabinet de « curiosités », sa bibliothèque ainsi que tous ses biens en argent. La ville décide alors d'ouvrir un nouveau musée enrichi des collections constituées lors des saisies révolutionnaires de 1792. Le musée est installé d'abord au couvent Saint-Martial, puis, pour faire face au volume croissant des collections, il est transféré en 1833 dans l'hôtel Villeneuve-Martignan, et inauguré en avril 1835. Aux œuvres offertes par de très nombreux donateurs (Esprit Requien, Edouard Raynolt, Marc Deydier, Noël Biret, Auguste-Louis Catelan, Joseph Rignault, Victor Martin et Marcel Puech) se sont ajoutées des acquisitions et des dépôts de l’État. Le musée est, jusqu’en 1984, géré avec la bibliothèque municipale. La séparation des deux institutions a ouvert depuis la possibilité d’un regroupement de toutes les œuvres du musée dans le seul hôtel de Villeneuve-Martignan. Musée municipal, il bénéficie du soutien d’un conseil d’administration dit Fondation Calvet, qui gère les revenus d’un patrimoine foncier important. Ce système, imaginé par Esprit Calvet en 1810, permet encore aujourd’hui l’acquisition d’œuvres d’art et la publication d’ouvrages scientifiques.
